U pacientov s idiopatickou z\u00e1palovou myopatiou je funkcia svalu ovplyvnen\u00e1 syst\u00e9mov\u00fdm z\u00e1palom nezn\u00e1mej etiol\u00f3gie, pri\u010dom porucha \u010dasto pretrv\u00e1va aj po potla\u010den\u00ed z\u00e1palu. Pravideln\u00e9 cvi\u010denie zlep\u0161uje energetick\u00fd metabolizmus a funk\u010dn\u00fd stav kostrov\u00e9ho svalu u zdrav\u00fdch \u013eud\u00ed u pacientov s diabetom 2. typu alebo idiopatickou z\u00e1palovou myopatiou. Cie\u013eom projektu je identifikova\u0165 \u0161truktur\u00e1lne, funk\u010dn\u00e9 a molekul\u00e1rne determinanty pozit\u00edvnych \u00fa\u010dinkov pravidelnej fyzickej aktivity na mitochondri\u00e1lnu respir\u00e1ciu, metabolick\u00fa substr\u00e1tov\u00fa preferenciu, ultra\u0161trukt\u00faru a kontraktiln\u00fa funkciu myot\u00fab. Ako model pou\u017eijeme vzorky kostrov\u00e9ho svalu ako aj z nich derivovan\u00e9 myotuby, z\u00edskan\u00e9 od metabolicky dobre charakterizovan\u00fdch pacientov pred a po ukon\u010den\u00ed tr\u00e9ningov\u00e9ho programu. Z\u00edskan\u00e9 poznatky bud\u00fa k\u013e\u00fa\u010dov\u00fdmi pre v\u00fdskum patofyziologick\u00fdch determinantov idiopatick\u00fdch z\u00e1palov\u00fdch myopati\u00ed.<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">K\u013e\u00fa\u010dov\u00e9 slov\u00e1:<\/span><\/h3>\n<p>cvi\u010denie, diabetes,idiopatick\u00e1 z\u00e1palov\u00e1 myopatia, \u013eudsk\u00fd kostrov\u00fd sval, metabolizmus, \u0161trukt\u00fara a funkcia myot\u00fab<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">Ciele:<\/span><\/h3>\n<p>Cie\u013eom tejto \u0161t\u00fadie je \u010do najkomplexnej\u0161ie definova\u0165 adapta\u010dn\u00fa odpove\u010f kostrov\u00e9ho svalu na pravideln\u00e9 cvi\u010denie u jedincov s chronick\u00fdm metabolick\u00fdm a\/alebo z\u00e1palov\u00fdm ochoren\u00edm. \u0160pecificky budeme \u0161tudova\u0165:<\/p>\n<ul>\n<li> Adapta\u010dn\u00e9 zmeny expresie k\u013e\u00fa\u010dov\u00fdch g\u00e9nov a obsahu pr\u00edslu\u0161n\u00fdch prote\u00ednov na cvi\u010denie v biopticky z\u00edskan\u00fdch vzork\u00e1ch kostrov\u00e9ho svalu od klinicky komplexne charakterizovan\u00fdch pacientov s idiopatickou z\u00e1palovou myopatiou a diabetes 2. typu a od zdrav\u00fdch dobrovo\u013en\u00edkov rovnak\u00e9ho veku a pohlavia. <\/li>\n<li> Mitochondri\u00e1lnu respir\u00e1ciu a hladinu koenz\u00fdmu Q10, esenci\u00e1lneho bioenergetick\u00e9ho faktora mitochondri\u00ed kostrov\u00e9ho svalu, a to vo vl\u00e1knach kostrov\u00e9ho svalu, ako aj v prim\u00e1rnych kult\u00farach \u013eudsk\u00fdch svalov\u00fdch buniek z\u00edskan\u00fdch od pacientov pred a po tr\u00e9ningu. <\/li>\n<li> Ultra\u0161trukt\u00faru svalov\u00fdch buniek elektr\u00f3novou mikroskopiou a obsah \u010di ko-lokaliz\u00e1ciu vybran\u00fdch prote\u00ednov, ktor\u00e9 definuj\u00fa funk\u010dn\u00fd stav diferencovan\u00fdch svalov\u00fdch buniek (myot\u00fab) STED fluorescen\u010dnou mikroskopiou. <\/li>\n<li> Adapta\u010dn\u00e9 zmeny excitability, kontraktility a v\u00e1pnikovej signaliz\u00e1cie diferencovan\u00fdch myot\u00fab. <\/li>\n<li> Hladiny t\u00fdch cirkuluj\u00facich a vo svale exprimovan\u00fdch bioakt\u00edvnych molek\u00fal,  ktor\u00e9 bud\u00fa experiment\u00e1lne regulovan\u00e9 v bunk\u00e1ch kostrov\u00e9ho svalu jedincov s chronick\u00fdm metabolick\u00fdm a z\u00e1palov\u00fdm ochoren\u00edm indukciou kontraktilnej aktivity. <\/li>\n<\/ul>\n<p>Hlavn\u00fdm cie\u013eom je identifikova\u0165 \u0161truktur\u00e1lne, funk\u010dn\u00e9 a molekul\u00e1rne determinanty \u00fa\u010dinkov fyzickej aktivity na kostrov\u00fd sval. Zameriame sa na mitochondri\u00e1lnu respir\u00e1ciu, metabolick\u00fa substr\u00e1tov\u00fa preferenciu, proces svalovej kontrakcie ako aj zmeny ultra\u0161trukt\u00fary svalovej bunky vyu\u017e\u00edvaj\u00fac vzorky kostrov\u00e9ho svalu  a diferencovan\u00e9 \u013eudsk\u00e9 myotuby od dobrovo\u013en\u00edkov s definovan\u00fdm fenotypom, z\u00edskan\u00e9 pred a po absolvovan\u00ed tr\u00e9ningov\u00e9ho programu.
