T\u00e1to regul\u00e1cia je naru\u0161en\u00e1 pri genetick\u00fdch ochoreniach CPVT-1 a ARVD-2, pri ktor\u00fdch RyR vykazuje mut\u00e1cie. Cie\u013eom projektu je ur\u010di\u0165 \u0161trukt\u00faru dvoch \u0161pecifick\u00fdch dom\u00e9n RyR z oblasti \u010dast\u00e9ho v\u00fdskytuCPVT\/ARVD mut\u00e1ci\u00ed (\u201edom\u00e9nov\u00fd prep\u00edna\u010d\u201c), d\u00f4le\u017eit\u00fdch z h\u013eadiska alosterickej regul\u00e1cie otv\u00e1rania kan\u00e1la a spresni\u0165 ich \u00falohu pri modul\u00e1cii pokojovej aktivity RyR. Ur\u010d\u00edme \u0161trukt\u00faru dom\u00e9n a ich vz\u00e1jomn\u00e9ho komplexu. Met\u00f3dami elektrofyziol\u00f3gie a konfok\u00e1lnej mikroskopie zist\u00edme \u00fa\u010dinok peptidov\u00fdch fragmentov odvoden\u00fdch zo sekvencie dom\u00e9n na pokojov\u00fa aktivitu izolovan\u00fdch RyR kan\u00e1lov ako aj RyR kan\u00e1lov in situ.
