V s\u00fa\u010dasnosti neexistuje dostupn\u00e1 efekt\u00edvna lie\u010dba, preto\u017ee ich molekul\u00e1rna podstata e\u0161te nie je dostato\u010dne pochopen\u00e1. Cie\u013eom projektu je \u0161t\u00fadium molekul\u00e1rnych mechanizmov arytmi\u00ed typu CPVT (katecholam\u00ednergn\u00e1 polymorfn\u00e1 komorov\u00e1 tachykardia), sp\u00f4soben\u00fdch mut\u00e1ciami RyR2 prote\u00ednu, ktor\u00fd riadi uvo\u013e\u0148ovanie v\u00e1pnika z vn\u00fatrobunkov\u00fdch z\u00e1sobn\u00edkov myocytov srdca. Z\u00e1kladn\u00e1 hypot\u00e9za projektu je, \u017ee CPVT arytmie vznikaj\u00fa n\u00e1hlym uvo\u013enen\u00edm v\u00e1pnika, ku ktor\u00e9mu doch\u00e1dza kv\u00f4li zmen\u00e1m vlastnost\u00ed RyR2 v d\u00f4sledku mut\u00e1ci\u00ed. Preto\u017ee s\u00fa n\u00e1znaky, \u017ee arytmie pozorovan\u00e9 u pacientov so zlyh\u00e1vaj\u00facim srdcom (z\u00edskan\u00e9 arytmie) s\u00fa tie\u017e zapr\u00ed\u010dinen\u00e9 chybn\u00fdm vn\u00fatrobunkov\u00fdm riaden\u00edm v\u00e1pnikovej homeost\u00e1zy a dysfunkciou RyR2, porovn\u00e1me mechanizmy zdeden\u00fdch arytmi\u00ed na modeloch transg\u00e9nnych zvierat vyzna\u010duj\u00facich sa mut\u00e1ciami RyR2 so mechanizmami z\u00edskan\u00fdch arytmi\u00ed na modeloch zvierat s hypertrofiou srdca a so zlyh\u00e1vaj\u00facim srdcom, ako aj na biopsi\u00e1ch \u013eudsk\u00e9ho myokardu. Pre lep\u0161ie pochopenie mechanizmu arytmi\u00ed, ako aj na predpovedanie n\u00e1chylnosti k vzniku arytmi\u00ed pou\u017eijeme matematick\u00e9 modelovanie. Kone\u010dn\u00fdm cie\u013eom projektov\u00e9ho t\u00edmu je vyvin\u00fa\u0165 nov\u00e9 n\u00e1vrhy pre diagnostiku a terapiu\u00e9 a testova\u0165 nov\u00e9 farmak\u00e1, ktor\u00e9 by mohli by\u0165 efekt\u00edvne v predch\u00e1dzani\u00ed vroden\u00fdm a z\u00edskan\u00fdm arytmi\u00e1m s\u00favisiacim s abnormalitami riadenia vn\u00fatrobunkov\u00e9ho Ca2+.<\/p>\n<p>Kolekt\u00edv Oddelenia pre v\u00fdskum svalov\u00fdch buniek \u00daMFG SAV sa podie\u013ea na projekte v troch oblastiach: prvou je charakteriz\u00e1cia vlastnost\u00ed mutovan\u00e9ho RyR2 prote\u00ednu a v spolupr\u00e1ci s ostatn\u00fdmi partnermi ur\u010denie zmien vlastnost\u00ed RyR2 zodpovedn\u00fdch za arytmogenicitu, druhou je testovanie interpret\u00e1cie experiment\u00e1lnych v\u00fdsledkov pou\u017eit\u00edm matematick\u00e9ho modelovania, tre\u0165ou je charakterizovanie mechanizmu \u00fa\u010dinku testovan\u00fdch antiarytmick\u00fdch liekov na aktivity RyR2.<\/td>\n<\/tr>\n<tr>\n<td
