V kardiomyocytoch doch\u00e1dza k n\u00e1rastu objemu kontraktiln\u00fdch myofibr\u00edl, vn\u00fatorn\u00e9ho membr\u00e1nov\u00e9ho syst\u00e9mu a mitochondrii. Cirkul\u00e1cia v\u00e1pnika v priebehu kontrak\u010dno &#8211; relaxa\u010dn\u00e9ho cyklu sa postupne internalizuje a vyv\u00edja sa diadick\u00fd syst\u00e9m zabezpe\u010duj\u00faci v\u00e4zbu excit\u00e1cie s kontrakciou prostredn\u00edctvom v\u00e1pnikovej signaliz\u00e1cie. Cie\u013eom projektu je detailne charakterizova\u0165 v\u00fdvoj uvo\u013e\u0148ovania v\u00e1pnika a v\u00fdvoj ultra\u0161trukt\u00fary di\u00e1d po\u010das postnat\u00e1lneho v\u00fdvoja myokardu potkana. Origin\u00e1lne \u00fadaje o v\u00fdvoji v\u00e1pnikovej signaliz\u00e1cie z\u00edskan\u00e9 na izolovan\u00fdch kardiomyocytoch pomocou laserovej skenovacej konfok\u00e1lnej mikroskopie v s\u00fa\u010dinnosti s met\u00f3dou patch-clamp budeme korelova\u0165 s \u00fadajmi o v\u00fdvoji ultra\u0161trukt\u00fary di\u00e1d z\u00edskan\u00fdmi pomocou elektr\u00f3novej mikroskopie. Z\u00edskan\u00e9 v\u00fdsledky umo\u017enia vysvetli\u0165 \u0161pecifik\u00e1 v\u00e1pnikovej signaliz\u00e1cie v kardiomyocytoch neonat\u00e1lneho srdca.<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">K\u013e\u00fa\u010dov\u00e9 slov\u00e1:<\/span><\/h3>\n<p>Myokard, ontogen\u00e9za, v\u00e1pnikov\u00e1 signaliz\u00e1cia, v\u00e4zba excit\u00e1cie s kontrakciou, konfok\u00e1lna mikroskopia, elektrofyziol\u00f3gia<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">Ciele:<\/span><\/h3>\n<p>Organiz\u00e1cia a funkcia jednotiek v\u00e4zby excit\u00e1cie s kontrakciou (di\u00e1d) svalov\u00fdch buniek myokardu u novorodencov je na perif\u00e9rii z\u00e1kladn\u00e9ho v\u00fdskumu i ke\u010f je dobre zn\u00e1me, \u017ee svalov\u00e9 bunky postnat\u00e1lneho myokardu podliehaj\u00fa prudk\u00e9mu v\u00fdvoju. Di\u00e1dy vznikaj\u00fa postnat\u00e1lne a rozv\u00edjaj\u00fa sa so zvy\u0161ovan\u00edm po\u017eiadaviek na pumpovaciu funkciu myokardu. Cie\u013eom tohto projektu je zisti\u0165 v\u00fdvoj funk\u010dnosti di\u00e1d z h\u013eadiska v\u00e1pnikovej signaliz\u00e1cie po\u010das transform\u00e1cie myocytov z embryon\u00e1lnej do termin\u00e1lne diferencovanej formy v priebehu dospievania myokardu. Vyu\u017eijeme na\u0161e origin\u00e1lne know-how na charakteriz\u00e1ciu v\u00e1pnikov\u00fdch sign\u00e1lov. Poznanie v\u00fdvoja \u0161trukt\u00fary a funkcie di\u00e1d umo\u017en\u00ed lep\u0161ie porozumie\u0165 \u0161pecifik\u00e1m funkcie vyv\u00edjaj\u00faceho sa myokardu novorodencov a farmakoterapie det\u00ed.
