V zdravom srdcovom myocyte doch\u00e1dza k mas\u00edvnemu uvo\u013eneniu v\u00e1pnika len po\u010das systoly, v d\u00f4sledku procesov vyvolan\u00fdch depolariz\u00e1ciou bunkovej membr\u00e1ny. Pri mnoh\u00fdch srdcov\u00fdch ochoreniach sa zvy\u0161uje uvo\u013e\u0148ovanie v\u00e1pnika po\u010das diastoly, \u010do m\u00e1 za n\u00e1sledok jednak pokles koncentr\u00e1cie v\u00e1pnika vo vn\u00fatrobunkov\u00fdch z\u00e1sobn\u00edkoch a teda zn\u00ed\u017een\u00fa schopnos\u0165 systolickej kontrakcie, jednak tzv. spont\u00e1nne diastolick\u00e9 uvo\u013enenie v\u00e1pnika, ktor\u00e9 vedie k oneskoren\u00fdm n\u00e1sledn\u00fdm depolariz\u00e1ci\u00e1m a arytmi\u00e1m. Vedeck\u00fdm z\u00e1merom projektu je objasni\u0165 molekul\u00e1rne mechanizmy regul\u00e1cie uvo\u013e\u0148ovania v\u00e1pnika po\u010das diastoly prostredn\u00edctvom ryanod\u00ednov\u00e9ho receptora (RyR) a vysvetli\u0165 d\u00f4sledky chybn\u00e9ho vr\u00e1tkovania RyR na homeost\u00e1zu v\u00e1pnika a v\u00e1pnikov\u00fa signaliz\u00e1ciu srdcov\u00e9ho myocytu. V\u00fdsledky projektu umo\u017enia identifikova\u0165 poruchy regul\u00e1cie uvo\u013e\u0148ovania v\u00e1pnika pri z\u00e1va\u017en\u00fdch srdcov\u00fdch ochoreniach.<br \/>\nPedagogick\u00fdm z\u00e1merom projektu je pripravi\u0165 jednu PhD \u0161tudentku pre \u010fal\u0161ie povolanie tak, aby mohla naplno rozvin\u00fa\u0165 svoj talent a dosiahnu\u0165 uplatnenie zodpovedaj\u00face jej schopnostiam. Po ukon\u010den\u00ed doktorandsk\u00e9ho \u0161t\u00fadia bude schopn\u00e1 za\u010da\u0165 samostatn\u00fa vedeck\u00fa kari\u00e9ru v b\u00e1date\u013eskom v\u00fdskume, alebo n\u00e1js\u0165 si uplatnenie v lek\u00e1rskom v\u00fdskume, v podnikate\u013eskej sf\u00e9re v oblasti \u201ehigh-tech\u201c technol\u00f3gi\u00ed, alebo sa sta\u0165 kvalitn\u00fdm vysoko\u0161kolsk\u00fdm pedag\u00f3gom.<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">K\u013e\u00fa\u010dov\u00e9 slov\u00e1:<\/span><\/h3>\n<p>V\u00e1pnikov\u00e1 homeost\u00e1za, v\u00e1pnikov\u00e1 signaliz\u00e1cie, ventrikul\u00e1rny myocyt, ryanod\u00ednov\u00fd receptor, elektrofyziol\u00f3gia, matematick\u00e9 modelovanie, srdcov\u00e9 ochorenia<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\"> Ciele:<\/span><\/h3>\n<p>Cie\u013eom projektu je charakterizova\u0165 molekul\u00e1rne mechanizmy regul\u00e1cie aktivity ryanod\u00ednov\u00e9ho receptora po\u010das diastoly a d\u00f4sledky por\u00fach t\u00fdchto mechanizmov na spont\u00e1nne uvo\u013enenie v\u00e1pnika po\u010das diastoly a na spriahnutie excit\u00e1cie s kontrakciou pri srdcov\u00fdch ochoreniach.
