Vz\u0165ah medzi molekul\u00e1rnym zlo\u017een\u00edm di\u00e1d a funk\u010dnos\u0165ou v\u00e1pnikovej signaliz\u00e1cie nie je dostato\u010dne objasnen\u00fd, nako\u013eko nie je pr\u00edstupn\u00fd priamej experiment\u00e1lnej manipul\u00e1cii. Vz\u0165ah \u0161trukt\u00fary a funkcie di\u00e1d by v\u0161ak malo by\u0165 mo\u017en\u00e9 sledova\u0165 v postnat\u00e1lnom obdob\u00ed v\u00fdvoja myokardu, kedy doch\u00e1dza k ich intenz\u00edvnej tvorbe a rastu. Cie\u013eom projektu je korelova\u0165 zmeny lok\u00e1lnej v\u00e1pnikovej signaliz\u00e1cie a prote\u00ednov\u00e9ho zlo\u017eenia a interakci\u00ed v jednotliv\u00fdch diadick\u00fdch komplexoch v priebehu ontogen\u00e9zy myocytov. Z\u00edskan\u00e9 \u00fadaje umo\u017enia formulova\u0165 hypot\u00e9zu o vz\u0165ahu \u0161trukt\u00fary a funkcie di\u00e1d na molekulovej \u00farovni a pom\u00f4\u017eu porozumie\u0165 adapt\u00edvnym procesom pri srdcovej hypertrofii na \u00farovni v\u00e1pnikovej signaliz\u00e1cie.<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">K\u013e\u00fa\u010dov\u00e9 slov\u00e1:<\/span><\/h3>\n<p>E-C v\u00e4zba, v\u00e1pnikov\u00e1 signaliz\u00e1cia, interakcia prote\u00ednov, diadick\u00e9 komplexy<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">Ciele:<\/span><\/h3>\n<p>Cie\u013eom projektu je charakterizova\u0165 priebeh tvorby a rastu diadick\u00fdch komplexov po\u010das dozrievania komorov\u00fdch myocytov z molekul\u00e1rno-biologick\u00e9ho h\u013eadiska v korel\u00e1cii s v\u00fdvojom E-C v\u00e4zby na \u00farovni jednotliv\u00fdch di\u00e1d. S\u00fastred\u00edme sa na prote\u00edny, ktor\u00fdch \u00faloha spo\u010d\u00edva v tvorbe a \u0161truktur\u00e1lnej stabiliz\u00e1cii diadick\u00e9ho komplexu (junktofil\u00edn, kaveol\u00edn), vo vlastnom prenose sign\u00e1lu medzi sarkolemou a intracelul\u00e1rnou membr\u00e1nou (DHPR a RyR kan\u00e1ly, Na\/Ca v\u00fdmenn\u00edk) a na prote\u00edny reguluj\u00face komunik\u00e1ciu medzi i\u00f3nov\u00fdmi kan\u00e1lmi sarkolemy a intracelul\u00e1rnej membr\u00e1ny (kalmodul\u00edn, junkt\u00edn, triad\u00edn a kalsekvestr\u00edn). Ot\u00e1zky, ktor\u00e9 si kladieme, s\u00fa:<\/p>\n<p>1.
