Wfs1 je exprimovan\u00fd vo vysokej miere v mozgu a srdci a je lokalizovan\u00fd v membr\u00e1ne endoplazmatick\u00e9ho retikula (ER), kde moduluje hladinu v\u00e1pnika a ER stres. Naviac, \u010das\u0165 pr\u00edznakov WS predstavuj\u00fa pr\u00edznaky typick\u00e9 pre mitochondri\u00e1lne ochorenia. T\u00fato skuto\u010dnos\u0165 podporuj\u00fa aj na\u0161e predbe\u017en\u00e9 v\u00fdsledky, kde potla\u010denie expresie Wfs1 v neur\u00f3noch malo za n\u00e1sledok zn\u00ed\u017eenie frekvencie f\u00fazi\u00ed a fragment\u00e1ciu mitochondri\u00ed. Hoci vysok\u00e1 expresia Wfs1 v srdci a srdcov\u00e9 komplik\u00e1cie u WS ned\u00e1vno zdokumentovan\u00e9 zd\u00f4raz\u0148uj\u00fa v\u00fdznam Wfs1 v srdci, pr\u00ed\u010dinou preva\u017enej v\u00e4\u010d\u0161iny \u00famrt\u00ed pacientov ost\u00e1vaj\u00fa neurologick\u00e9 komplik\u00e1cie. Ako je mo\u017en\u00e9, \u017ee mut\u00e1cie Wfs1, ktor\u00e9 u pacientov sp\u00f4sobuj\u00fa z\u00e1va\u017en\u00e9 po\u0161kodenie mozgov\u00fdch funkci\u00ed sa neprejavia aj kardi\u00e1lnymi pr\u00edznakmi? Jedn\u00fdm z mo\u017en\u00fdch vysvetlen\u00ed by mohlo by\u0165, \u017ee v srdci existuje kompenza\u010dn\u00fd mechanizmus, ktor\u00fd nie je pr\u00edtomn\u00fd v neur\u00f3noch.<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">K\u013e\u00fa\u010dov\u00e9 slov\u00e1:<\/span><\/h3>\n<p>mitochondria, endoplazmatick\u00e9 retikulum, Wolframov syndr\u00f3m, f\u00fazia-delenie, v\u00e1pnik, kontraktilita<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">Ciele:<\/span><\/h3>\n<p>Cie\u013eom predkladan\u00e9ho projektu je objasni\u0165, \u010di u transg\u00e9nnych my\u0161\u00ed s del\u00e9ciou Wfs1 g\u00e9nu s\u00fa pr\u00edtomn\u00e9 poruchy kontraktility srdca na org\u00e1novej aj bunkovej \u00farovni. \u010ealej, ak Wfs1 v\u00fdznamne ovplyv\u0148uje dynamiku a morfol\u00f3giu mitochondri\u00ed v neur\u00f3noch, je d\u00f4le\u017eit\u00e9 uk\u00e1za\u0165, \u010di tento jav je pr\u00edtomn\u00fd aj v kardiomyocytoch. Na vysvetlenie javu siln\u00e9ho vplyvu ER-viazan\u00e9ho prote\u00ednu na mitochondrie chceme presk\u00fama\u0165, \u010di tento jav je sp\u00f4soben\u00fd tesnej\u0161ou interakciou ER a mitochondri\u00ed (pomocou elektr\u00f3novej mikroskopie) a\/alebo je podkladom v\u00e1pnikov\u00e1 signaliz\u00e1cia (konfok\u00e1lna mikroskopia).<\/p>\n<p>Hlavn\u00e1 hypot\u00e9za: Ak\u00fd mechanizmus kompenzuje nedostatok Wfs1 prote\u00ednu v kardiomyocytoch v pr\u00edpade del\u00e9cie Wfs1 g\u00e9nu? Mohol by sa tento ochrann\u00fd mechanizmus da\u0165 aktivova\u0165 aj v neur\u00f3noch a napom\u00f4c\u0165 tak pri lie\u010dbe neurodegenerat\u00edvnych ako aj kardiovaskul\u00e1rnych chor\u00f4b?<\/p>\n<p>Cie\u013e 1: Je naru\u0161en\u00e1 mitochondri\u00e1lna dynamika vo Wfs-\/- myocytoch?<br \/>\nCie\u013e 2: Sp\u00f4sobuje deficiencia Wfs1 negat\u00edvne zmeny v ultra\u0161trukt\u00fare mitochondri\u00ed?
