Existuje mno\u017estvo d\u00f4kazov, \u017ee aj ke\u010f \u017eeny v predmenopauz\u00e1lnom veku maj\u00fa ni\u017e\u0161ie riziko KVO v porovnan\u00ed s mu\u017emi rovnak\u00e9ho veku, toto sa vytr\u00e1ca u diabetick\u00fdch premenopauz\u00e1lnych \u017eien, kde pre KVO dominuje diabetick\u00e1 kardiomyopatia (DCM). DCM sa vyzna\u010duje \u0161truktur\u00e1lnymi ako aj funk\u010dn\u00fdmi zmenami myokardu. Preva\u017en\u00e1 v\u00e4\u010d\u0161ina hum\u00e1nnych ako aj experiment\u00e1lnych \u0161t\u00fadi\u00ed vr\u00e1tane modelu potkanov Zucker Diabetic Fatty (ZDF), sa v\u0161ak doposia\u013e s\u00fastredila na \u0161t\u00fadium DCM u mu\u017eov\/samcov, kde konkr\u00e9tne u samcov potkanov ZDF bola pozorovan\u00e1 diastolick\u00e1 dysfunkcia, porucha v\u00e1pnikovej signaliz\u00e1cie a hypertrofia myocytov. Preto je na\u0161\u00edm cie\u013eom charakterizova\u0165 vn\u00fatrobunkov\u00fa dynamiku v\u00e1pnika a ultra\u0161trukt\u00faru kardiomyocytov u sam\u00edc potkanov ZDF a porovna\u0165 ich charakteristiky v \u0161t\u00edhlych a ob\u00e9znych modeloch s vyvinutou DCM alebo bez nej, aby sme rozl\u00ed\u0161ili mechanizmy sp\u00f4sobuj\u00face patol\u00f3giu vyvolan\u00fa di\u00e9tou, obezitou a cukrovkou.<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">K\u013e\u00fa\u010dov\u00e9 slov\u00e1:<\/span><\/h3>\n<p>kardiomyopatia, diabetes, obezita, vysokotukov\u00e1 strava, v\u00e1pnikov\u00fd tranzient, elektr\u00f3nov\u00e1 mikroskopia<\/td>\n<\/tr>\n<tr>\n<td>\n<h3><span class=\"H3\">Ciele:<\/span><\/h3>\n<p>Cie\u013eom tohto projektu je charakterizova\u0165 kardiomyocyty sam\u00edc potkanov ZDF, najm\u00e4 ich intracelul\u00e1rnu v\u00e1pnikov\u00fa dynamiku a ultra\u0161trukt\u00faru, a porovna\u0165 ich medzi \u0161t\u00edhlymi (fa\/+) a ob\u00e9znymi (fa\/fa) modelmi s alebo bez diabetickej kardiomyopatie (DCM) vyvolanej vysokotukovou stravou (HFD), aby sme rozl\u00ed\u0161ili mechanizmy sp\u00f4sobuj\u00face patologick\u00e9 zmeny vyvolan\u00e9 stravou, obezitou a cukrovkou.<\/p>\n<p>Prvou hypot\u00e9zou pre podporu tohto cie\u013ea je, \u017ee kardiomyopatia (CM) u sam\u00edc, rozv\u00edjaj\u00faca sa v skupine fa\/fa HFD, je v\u00fdsledkom obezity a\/alebo HFD, a nie samotnej hyperglyk\u00e9mie. Na potvrdenie tejto hypot\u00e9zy budeme sk\u00fama\u0165 srdcov\u00fa patol\u00f3giu u nediabetick\u00fdch zvierat, t.j. normoglykemick\u00fdch ob\u00e9znych sam\u00edc na \u0161tandardnej strave (fa\/fa StD) a chud\u00fdch sam\u00edc na strave s vysok\u00fdm obsahom tukov (fa\/+ HFD). Budeme analyzova\u0165 fenotyp srdca vo v\u0161etk\u00fdch \u0161tyroch skupin\u00e1ch sam\u00edc potkanov, t.j. fa\/fa HFD, fa\/fa StD, fa\/+ HFD a fa\/+ StD (obr. 1 A). O\u010dak\u00e1vame v\u00fdznamn\u00fd rozdiel medzi srdcov\u00fdm fenotypom v kontrolnej skupine fa\/+ StD (t. j. absencia CM) a fenotypom v skupine fa\/fa HFD (pr\u00edtomnos\u0165 DCM). Ak sa CM prejav\u00ed v skupine fa\/+ HFD, ale nie v skupine fa\/fa StD, bude to indikova\u0165 siln\u00fd \u00fa\u010dinok HFD na srdcov\u00fa patol\u00f3giu. Naopak, ak bude CM pr\u00edtomn\u00e1 v fa\/fa StD, ale nie v skupine fa\/+ HFD, bude to znamena\u0165, \u017ee CM je vyvolan\u00e1 obezitou. Ak ani v jednej zo skup\u00edn fa\/fa StD a fa\/+ HFD CM nebude pr\u00edtomn\u00e1, nazna\u010duje to \u0161pecifick\u00fd vn\u00fatorn\u00fd mechanizmus, ktor\u00fd sa vyv\u00edja len vtedy, ke\u010f s\u00fa pr\u00edtomn\u00e9 v\u0161etky faktory (HFD, obezita a diabetes).<\/p>\n<p>Druh\u00e1 hypot\u00e9za predpoklad\u00e1, \u017ee HFD je potrebn\u00e1 pre pretrv\u00e1vanie DCM u sam\u00edc potkanov fa\/fa ZDF. Diabetick\u00fd stav, teda hyperglyk\u00e9mia a inzulinop\u00e9nia, u sam\u00edc fa\/fa pretrv\u00e1va po \u00faplnom rozvinut\u00ed cukrovky aj po prechode na StD. Ak HFD nie je potrebn\u00e1 na udr\u017eanie DCM, fenotyp DCM by sa mal \u010falej rozv\u00edja\u0165 aj po prechode diabetick\u00fdch sam\u00edc na SfD.
